magiaさんは合っていますよ。 「４日間休み」という重要な点がわかっているのですから、始めの日を聞けば大丈夫ですよね。 もし「４日間」がわからなかった場合。 「when does your vacation start?」 あなたの休みはいつから始まるの？ 「how long is it?」 どれくらい長いの？ を連続で書けばemailの場合いつからいつまでと相手も返してくれるはずです。 もしいつ始まるかわかっている場合 「how long is your vacation going to be」 あなたの休みはどれくらい長くなるの？ と聞けばいいと思います。 和訳すると何だかぎこちないですが、英語だけ読むととてもさりげないので目上の方にも友達にも使えますよ。

いろいろな言い回しを考えて、なおかつ誤解の心配が残るくらいなら、 Do you mean your vacation will be Friday through Monday? のように、バッチリ尋ねるのがいいと思います。もちろん、曜日だけで不安ならdateも入れて。

4日間とわかっているので、いつ始まるかを聞くだけでOKです。これから先の予定を聞くので、 When will your（必要ならばsummer)vacation start? で大丈夫です。

口語なら"from when to when?" "from when?"で通じますよ。 ４日間と答えが出ているので、初めの日にちを聞くだけでいいと思います。
