Is that OK? は、 「それでいいかな」（今風に言えば）「それで良かったかな？」、 あるいは単純に「それでOK?」、という意味で、 いろんな場面で気軽に使えるフレーズです。 Hope you are OK では、 I を省略していて、I hope you are OK ということだと思います。 よく気軽なemailの中で使ったりします。 意味は簡単で、「（あなたが）元気だといいけど」、「元気だよね」 という感じです。 ほかの方の回答も参考にして、ニュアンスをつかんで下さいね。

Is that ok? は「それでいい？」という意味です。 Hope you are ok. は「君が大丈夫（元気）だといいんだけど」という意味です。 しかし Is that ok? と Hope you are ok. が並んで書いてあったなら、 「それでいい？」「（君が）それでいいこと願ってるよ」というふうになるでしょう。
