In 1952, two scientists, Robert Briggs and Thomas King, removed the nucleus from the cell of one frog, and put it into an egg cell of another frog whose nucleus had been removed. The end result: A cloned frog. The inserted nucleus was taken from an “unprogrammed cell” of an embryo. Unprogrammed cells of an embryo can turn into any kind of cell to form, for example, bones, blood, a heart, or a kidney. Briggs and King thought that only nuclei from embryos, not adult cells, could be used to create clones. They believed that adult cells, such as skin cells that are programmed to grow new skin over a wound, could only create new cells of the exact same type. (1) to formは直前のany kind of cellを修飾する形容詞用法の不定詞。any kind of cellは、意味上不定詞to formの意味上の主語の働きをする。 (2) to formは前のturn (into any kind of cell)を修飾する副詞的用法の不定詞。

To readily calculate some of the physical characteristics of crystals, such as atomic density and density of three electrons, we must know the number of lattice points per cell. You may think that such a determination is quite trivial, but it is surprising how many people have difficulty with it! The number of lattice points per cell, N, is given by the equation N= Ni+ Ni/2+ Nc/8 Where Ni is the number of lattice points in the interior of the cell (these points “belong” only to one cells), Nf is the number of lattice points on faces (these are shared by two cells), and Nc is the number of lattice points on corners (these are shared by eight cells). For the three cubic unit cells the number of lattice points per cell is Primitive cubic (cubic P) 1 Body-centered cubic (cubic I) 2 Face-centered cubic(cubic F) 4 All primitive cells have one lattice point per cell. All nonprimitive cells have more than one lattice point per cell.

(1)Embryonic stem (ES) cells derive from totipotent cells of the early postimplantation embryo and are capable of unlimited, undifferentiated proliferation in vitro while maintaining the potential to form cell types of all three germ layers. (2)The bone marrow compartment is largely made up of committed progenitor cells, noncirculating stromal cells that have the ability to develop into mesenchymal lineages (termed mesenchymal stem cells), and HSCs. (3)However, reports of participation of hematopoietic cells in neurogenesis, the conversion of adult neural stem cells into hematopoietic cells, and the demonstration that adult mouse neural stem cells can give rise to cells of all germ layers seemed to confirm that differentiation could occur out with the original germ layer. This challenged the conventional trilaminar view of organ development may exhibit as much pluripotentiality as ES cells. (4)Examination of the host liver demonstrated hepatic cells that were donor derived, as detected by expression of dipeptidyl peptidase IV enzyme (DPPIV+) in DPPIV– rats or the Y chromosome in female animals. (5)The significance of such a survival advantage is confirmed in a transgenic model based on the protective effect of the antiapoptotic gene, Bcl-2, against Fas-mediated cell death. Bone marrow from mice expressing this transgene, under the control of a liver-specific promoter, was infused into normal mice. Some mice underwent repeated injections with a Fas-agonist antibody to induce liver injury, whereas others did not. Only those that had received antibody injections showed mature hepatocytes expressing Bcl-2, implying that transdifferentiation is inefficient under physiological conditions and that tissue injury such as accumulation of toxic catabolites or apoptotic challenge is required to generate a more robust response.

How progenitor cells decide their fate is a question that underlies all of developmental biology but is poorly understood. While complex regulatory networks are known to maintain cells in distinct cell fates ( [Davidson et al., 2002] , [Novershtern et al., 2011] and [Odom et al., 2004] ), we know little about how cells integrate signals and reorganize these networks to allow fate transitions.

Principles of neuronal developmentの Counting Cells p132-133を読みました。 (1) It might seem a straightforward matter to assess by accurate cell counts the degree of neuronal degeneration during the development of a particular region of the nervous system. However, this information is actually quite hard to come by, and a number of acrimonious disputes about neuronal death have revolved around methodology. (2)Counting cells in a central nucleus is relatively simple as long as the nucleus remains discrete throughout the developmental period of interest. (3)but this method suffers from the assumption that developing cells take up the marker equally well at different developmental stages and that diffusion of the marker is equally restricted in small and large targets. (4)Counting neurons in sections requires correction for "double counting", that is for counts of the same cell in two or more adjacent sections.

A remarkble feature of most viruses is the symmetrical structure of their prptein cost, built up of one or more subunits packed in a way that recalls a chemical crystal more than a from of life.Figure 2.3 illustrates two kinds of symmetry favoured by viruses.Viruses unfortunately do not lend themselves to the branched system of classification used for most animals, and are usually classified on the basis of their mncleic acid and whether or not they posses a lipid envelope outside their protein cost. This envelope is acquired by the virus from the host cell in the process of making its exit-a rpocess known as budding which enables the virus to sruvive outside the cell sufficiently long, for example,to spread elsewhere via the bloodstream. Whethaer a particular virus spreads in this way or directly from one cell to its immediate neighbour has a considerrable bearing on both the pattern of infection and the development of immunity. Thus an enveloped virus can leave its host cell without destroying it,while the non-enveloped sort will rupture the cell (`cytolysis`);the latter are known as cytopathic viruses because of their ability to damage cells and tissues, that is,cause pathology.Figure 2.4 illustrates these two means of spread.

(3)The selectivity of these treatments depends on quantitative differences in the rates of division between cancer and normal cells; relative levels of the direct molecular target (such as the DNA topoisomerases); and tumour defects in pathways that are related to the target of treatment, such as in checkpoint signalling pathways (including the p53 pathway). (4)This discovery of the cellular DNA-damage response was made many years ago when it was observed that DNA damage, such as that caused by ionizing radiation (IR), temporarily inhibits cell-cycle progression, and that the delay period provides the time needed to repair the DNA and reduces the lethal effect of radiation. (5)These include inhibition of cell-cycle progression; activation of DNA repair; maintenance of genome stability; and, in some cell types or when damage is beyond repair, initiation of permanent cell-cycle blockade (cellular senescence), or elimination of such cells by cell death. So, the narrower definition of the checkpoint pathway is not adequate to explain its overall functionality, and the cell-cycle checkpoint pathways as such represent only some aspects of the global cellular network that is triggered in response to DNA damage.
